A Comparative Study of the Separation of Oleanolic Acid and Ursolic Acid in Prunella vulgaris by High-Performance Liquid Chromatography and Cyclodextrin-Modified Micellar Electrokinetic Chromatography

چکیده
A high-performance liquid chromatographic (HPLC) method and a cyclodextrin-modified micellar electrokinetic chromatographic (CD-MEKC) method were developed to separate and determine oleanolic acid (OA) and ursolic acid (UA) in Prunella vulgaris. HPLC separations were carried out on a Hedera ODS C18 column with methanol -H2Oacetic acid (85:15:0.3, v/v/v) as mobile phase at a flow-rate of 0.8 ml min. CD-MEKC analysis was performed on a CL1030 capillary electrophoresis system with a 6% (v/v) methanol solution (pH = 9.0) containing 10 mM disodium tetraborate, 10 mM sodium dihydrogen phosphate, 50 mM sodium dodecylsulfate (SDS), 15 mM 2-hydroxypropyl-β-cyclodextrin (2-HP-β-CD) as background electrolyte. The analytical results of HPLC and CD-MEKC were compared with each other. CD-MEKC has better analytical efficiency for two components, and the analytical time (15 min) was shorter than that of HPLC (35 min).

Prunella vulgaris L. (Labiatae) contains a variety of structurally diverse natural products, primarily rosmarinic acid (RA), ursolic acid (UA) and oleanolic acid (OA), which possess a wide array of biological properties. In the present study, P. vulgaris was harvested at three developmental stages (vegetative, full-flowering and mature-fruiting stages), dissected into stem and leaf tissues and ...
متن کاملChanges in bioactive components related to the harvest time from the spicas of Prunella vulgaris.
CONTEXT Prunella vulgaris L. (Labiatae) is a perennial plant common in China and Europe and is rich in rosmarinic acid (RA), ursolic acid (UA), and oleanolic acid (OA). The dried spica of P. vulgaris has been used as traditional medicine in China for over a hundred years. To our best knowledge, no study has been conducted to determine the influence of harvesting time on concentrations of bioact...
متن کاملCombination of active components enhances the efficacy of Prunella in prevention and treatment of lung cancer.
The efficacy of Prunella extracts in the prevention and treatment of lung cancer has been attributed to different components. In this study, an "active components combination model" hypothesis was proposed to explain the anti-tumor activity of Prunella. The efficacy of Prunella extracts from different regions was compared in vitro and in vivo, and the TNF-α activity in serum of tumor-bearing mi...

Prunella vulgaris L. (PV) has been used as a herb for chemoprevention of lung cancer. In this study, the main active compound, oleanolic acid (OA) was isolated from an ethanol extract and its chemical structure was identified according to the results of high performance liquid chromatography (HPLC), high performance thin layer chromatography (HPTLC) and liquid chromatography-mass spectrography ...
متن کاملSeparation of Sedative – Hypnotic Drugs with Mixed Micellar Liquid Chromatography
Separation of ten sedative- hypnotic drugs was performed by RP-HPLC using mixed micellar mobile phase. Effect of temperature, type and amount of organic modifier in mobile phase on efficiency (N) and asymmetry factor (B/A) showed that, the appropriate conditions for a good separation were 35°C and 7% (V/V) butanol in mobile phase. Variations of selectivity factor versus butanol concent...
